Resazurin microtiter assay for detection of drug resistance and determination of critical concentration of cycloserine resistance of Mycobacterium tuberculosis

چکیده
We evaluated the feasibility of resazurin microtiter assay (REMA) for detecting cycloserine resistance of Mycobacterium tuberculosis. The cutoff value of the critical concentration of cycloserine resistance in vitro about M. tuberculosis isolates was determined innovatively by drawing the receiver operating characteristic (ROC) curve for the first time. The spending time of REMA ranged from 6 to 9 days. The ROC curve analysis showed that the area under the curve (AUC) was 0.950, and P value was <0.001. The critical concentration of cycloserine in vitro was >16 μg/ml based on the ROC curve. At this point, the sensitivity of REMA was 79.17%, and the specificity was 100% when compared to the proportion method (PM). Resazurin method was found to be simple, reliable and inexpensive to perform for the rapid detection of anti-tuberculous drugs.

Resazurin microtiter assay plate: simple and inexpensive method for detection of drug resistance in Mycobacterium tuberculosis.
A method for detecting multidrug-resistant Mycobacterium tuberculosis by using a reduction of resazurin is described. Eighty clinical isolates were evaluated against isoniazid and rifampin; results at 7 days were compared with those of the proportion method. Specificity and sensitivity were excellent. The method is simple, inexpensive, and rapid and might be used with other antituberculosis drugs.

Tuberculosis (TB) ranks second, next to AIDS making it most formidable disease if the present age. One of the crucial enzymes involved in cell wall synthesis of Mycobacterium tuberculosis, InhA (enoyl acyl carrier protein reductase) has been authenticated as an effective target for anti-mycobacterial drug development. In the current work, we have developed novel derivatives of 1,2,4-triazole-5-...

The potentially pathogenic Non-Tuberculosis Mycobacteria (NTM) are emerging nowadays which result in pulmonary and non-pulmonary infections in human. This group of bacteria consists of at least 200 different species. While the pulmonary disease is the most common form of NTM infections, NTM can cause diffused infections as well as extrapulmonary infections in every organ, such as bone marrow, s...

Drug resistance (especially multiple drug resistance) in Mycobacterium tuberculosis makes global concerns in treatment and control of tuberculosis. Rapid diagnosis of drug resistant strains of the bacteria has vital importance in the prognosis of the disease. The aim of this study was to identify the mutations responsible for drug resistance in Mycobacterium tuberculosis strains derived from pa...
